IGF-1 LR3 is a recombinant 83-amino-acid analog of human insulin-like growth factor 1. It differs from native IGF-1 by two structural modifications: an arginine substitution for glutamic acid at position 3, and a 13-amino-acid N-terminal extension peptide. Together these changes markedly reduce the molecule's affinity for IGF-binding proteins (IGFBPs), which in laboratory systems is associated with a longer functional half-life and increased potency at the IGF-1 receptor relative to unmodified IGF-1. In published research, IGF-1 LR3 is widely used as a reference ligand for studying IGF-1R signaling, cellular proliferation, and protein-synthesis pathways in cell-culture models.
