NA-Semax is an N-acetylated analog of Semax, a synthetic peptide combining a fragment of adrenocorticotropic hormone (ACTH 4–7) with a Pro-Gly-Pro stabilizing tripeptide. The parent sequence (Met-Glu-His-Phe-Pro-Gly-Pro) is a non-hormonal ACTH fragment — it retains the neuromodulatory portion without the corticotropic activity of the full hormone. N-terminal acetylation is a further modification associated in laboratory settings with increased peptidase resistance and improved stability relative to unmodified Semax. In published research, Semax and its analogs are used as tool compounds for investigating BDNF and NGF expression, dopaminergic and serotonergic signaling, and enkephalin-degrading enzyme activity in cell-culture and animal model systems.
